Estimation of Damage to Human Spermatozoa During Cryopreservation by measurement of Release of Glutamic-Oxalacetic Transaminase

Abstract

|
Release of glutamic-oxalacetic transaminase(GOT) has been used as an indicator of cell damage during cryopreservation of spermatozoa from domestic animals. This study was planned to detemine whether changes in seminal GOT activity could be
measured
after cryopresxervation of human spermatozoa and to correlate the findings with the extent of damage detected with eosin staining. Seminal GOT correlated positively with sperm density, and increases were measured after cryoperservation. The
amount
of
GOT released was not influenced by the speed of cryopreservation(fast vs. Slow )and reflected the extent of damage to the plasma membrane of the spermatozoa seen by light microscopy. Measurement of GOT may be a useful indicator of the cellular
integrity
of cryopreserved spermatozoa.
